GILL'S HEMATOXYLIN

Hematoxylin

Gill's hematoxylin is good dye for nuclear staining. It can be combined with eosin. Compared with other hematoxylins, Gill's hematoxylin gives more stable and reproducible staining. The nucleoli are well-stained. Since it does not contain mercury or alcohol, it can be combined with other techniques, such as immunohistochemistry.

Procedure

750 ml distilled water

250 ml ethylene glycol

2 g hematoxylin (C.I. 75290)

0.,2 g sodium iodate

17,6 g aluminium sulfate

20 ml glacial acetic acid

Preparation

The components are added following the order describe and the solution is stirred for about 1 h. The solution can be used immediately after.

It can be store longer than other hematoxylins.

The amount of hematoxylin, sodium iodate and aluminium sulfate can be increased to make the staining stronger. However, the proportions of the three elements should remain constant. For example: 4 g, 0.4 g, 40 g, or 6 g, 0.6 g, 80 g. The other components of the solution are not changed.

When high concentrations of hematoxylin, sodium iodate and aluminium sulfate are used, the final solution can be diluted.
