Cresyl violet is a basic synthetic dye extensively used to stain the nervous tissue, although it is also suitable for staining mastocytes and cartilage. Cresyl violet is different from crystal violet, which is for bacteria Gram staining. In the nervous tissue, cresyl violet can be combined with luxol fast blue for myelin staining.
Procedure (a)
0,1 g cresyl violet
100 ml distilled water
1 or 2 (about 0,25 ml) drops of acetic acid
Procedure (b)
0,6 ml acetic acid in 100 ml of distilled water (A)
1, 36 mg sodium acetate in 100 ml of distilled water (B)
Acetate buffer: mix solutions A:B (9:1)
1 g cresyl violet in 100 ml distilled water
Working solution ... Acetate buffer: 1 % cresyl violet (1:1)
Notes
In the procedure B, adjust the pH of solution B to 3,7 before mixing with cresyl violet.
