CARAZZI'S HEMATOXYLIN

Hematoxylin

Carazzi's hematoxylin is a good dye to stain nuclei, and with few background staining. It can be used for general staining, combined with eosin. Carazzi's hematoxylin staining is a progressive staining.

Components

800 ml distilled water

200 ml glycerol

1g hematoxylin (C.I. 75290)

0,2g Sodium Iodate

50 g Potassium alum (KAl(SO4)2·12H2O)

Procedure

Mix hematoxylin and glycerol. Solve the sodium iodate in a bit of water and dissolve the potassium alum in the remaining water. Mix the hematoxylin-glycerol and the potassium alum, and then slowly add the sodium iodate. Filter the final solution.

The solution can be used immediately, and it is stable for months.

Notes

It is a progressive statining, with low background.

In frozen sections, a double staining is recommended.
